The detection of genetically modified organisms is often pursued by the polymerase chain reaction (PCR). The detection includes basically three steps: DNA extraction from the sample, PCR with detection of products, and data evaluation. In the presented report a suitable protocol for the DNA isolation was searched for. From the published methods, five were chosen and used for the isolation of the DNA from wheat, soybean, potatoes and corn. The methods were evaluated by means of different parameters, likethe DNA purity, amplifiability by PCR, cost and duration of the procedure. The modified CTAB method and DNeasyTM Plant Mini Kit were found the most suitable methods of extraction. Then the selection of optimal conditions for detection of transgenic wheat and transgenic potato was performed. The samples were tested for possible occurrence of sequences of transgenic DNA that are used in commonly available genetically modified products. The most frequent sequences, regulating expression of
