Flow cytometric analysis of Saccharomyces cerevisiae

Two methods of preservation and storage of yeast cultures are tested. Freest yeast population suspended at medium containing glycerol (5% v/v) as a cryoprotectant and periodic sub-culture using agar slopes (slants). This work was carried out with three widely used Czech lager strains - No. 2, 7 and 95. All yeasts were analyzed by flow cytometry prior to storage and after storage. Fluorescent intensity of DNA was analysed by flow cytometry and signal was used as a marker for cell cycle determination. During experiments were monitored changes in significant properties of yeast (glycogen, proteins, DNA) by fluorescent methods. It seems that the impact of storage conditions has not influence on basic characteristic of yeast population, dynamic of microbialgrowth, cell cycle and technological properties of yeast cultures.
