Immunoaffinity chromatography (IAC) or immunoextraction (IE) is based on a highly selective and affinitive interaction between antibodies immobilized on a solid support and analytes-antigens present in a sample. Thus enabling to concentrate and clean-upthe analytes from complex biological matrixes, such as plant extracts. Isoflavonoids belong to secondary plant metabolites and some of them are known for their pharmacological activities such as estrogenic, antiviral, antibacterial, antioxidative etc. Legumes are known as their main source. Nevertheless, the presence of isoflavonoids has been reported in about sixty other families. In non-legumes, isoflavonoids are present in lower concentrations, which results in a more difficult detection and identification. This was the reason for the development of an off-line combination of IAC and HPLC-MS for the detection of isoflavonoids in non-legume plant extracts. Immunosorbents were prepared by immobilization of polyclonal rabbit antibodies
