Isoflavonoids are plant phenolics known for their multiple pharmacological activities such as estrogenic, antibacterial, antioxidative etc. While legumes are their main source, the presence of isoflavonoids has been reported in about fifty other families. In non-legumes, isoflavonoids may be present in low concentrations, which complicates their detection and identification in complex biological matrixes. Two methods of sample clean-up prior to HPLC-ESI-MS, i.e. SPE (Solid Phase Extraction) and IAC (Immunoaffinity Chromatography), were compared. Conventional C18 reversed-phase sorbent cartridges (Waters Oasis, 30 ?m) were used for SPE. Immunosorbents for IAC were prepared by immobilisation of polyclonal rabbit antibodies against isoflavonoid-BSA (i.e.genistein, biochanin A and daidzein) conjugates on Affi-Gel 10 solid support (Bio-Rad Laboratories).
