    Influence of various enviromental factors on phytochemical profiles of agrimony (Agrimonia eupatoria L.)

    As a herb used in traditional medicine, Agrimonia eupatoria L. contains many bioactive compounds, such as flavonoids, phenolic acids and tannins. These compounds from the group of polyphenols have number of positive effects (e.g. antioxidant activity, antimicrobial activity) on human health. Because of that, the popularity of herbal extracts is nowadays growing and they find multiple applications. However, various factors can influence the profiles of bioactive compounds in the plant and through that efficacy of plants extract. This study was performed to unravel the impact of selected factors (variety, location, growing phase) on the agrimony phytochemical composition focusing mainly on polyphenols. An extensive set of agrimony samples from different locations of Czech Republic, differently processed (dried, fresh) and picked in various vegetation phases were extracted with 40% (v/v) ethanol and analyzed with ultra-high-performance liquid chromatography coupled with high-resolution tandem mass spectrometry (UHPLC-HRMS/MS). Multidimensional statistical methods such as principal component analysis (PCA) and partial least-squares discriminant analysis (PLS-DA) were employed to visualize the differences among the samples, followed by markers identification for the observed clustering with agrimony variety and growing locality as the most important factors. Targeted screening of polyphenolic compounds against the created in-house spectral library was also carried out and differences in presence of some individual polyphenols among the samples were observed. Additionally, trends in the content of bioactive compounds during the plant growing season were examined in the untargeted data as well as for the identified polyphenols through PatternHunter software. Trends in the ratios of flavonoid aglycones and their glycosides were also examined with the highest ratio of glycosides in the phase where agrimony begins to bloom. The results of this study are highlighting the variability of plants intended for use in medicinal preparations and the need for awareness of growing and harvesting conditions of medicinal plants.
