Introduction: Inflammatory bowel diseases (IBD), a group into which mainly Crohn’s disease and ulcerous colitis belong, are commonly spread through the world. A new option for treatment of IBD by using natural products is targeted in this project. Glucan particles are used as a vehicle, since β-glucan can interact with immunity system in organism. The hollow glucan particles, containing an active substance, e.g. natural anti-inflammatory drugs, can deliver their content into the targeting area and increase its bioavailability. Anti-inflammatory drugs naturally occurring in plants are chosen with limited solubility in water. In this project curcumin was tested due to its anti-inflammatory activity. Methodology: Glucan particles were prepared from Saccharomyces cerevisiae (baker’s yeast). The loaded microcapsules were prepared by mixing a solution of curcumin with pure glucan particles and subsequently removing the solvent by controlled evaporation process. The anti-inflammatory activity was tested in vitro on macrophage-like cells and in vivo in rats. Results: Pure glucan particles with properties suitable for microencapsulation were prepared from native yeast. A method for loading curcumin into glucan particles with required loading efficiency was developed. The anti-inflammatory activity of prepared microparticles with curcumin was tested on macrophage-like cells and in male Wistar rats. Conclusion: The prepared glucan particles from baker´s yeast were used as vehicle for an anti-inflammatory natural substance - curcumin. This formulation targets the inflammatory area in guts because glucan particles containing an active substance can be phagocytosed by immunity cells in Peyer’s patches present in the guts. Glucan particles are a promising delivery system, especially for diseases connected with the immune system, and can be used in treatment of IBD in the future.
