The study reports the successful cloning and functional characterization of the wheat powdery mildew resistance gene Pm6, originating from Triticum timopheevii. Using map-based cloning, chromosome sorting, EMS mutagenesis, and transcriptomic analysis, the authors identify TraesLAC2B03G00988960 as the gene underlying Pm6, encoding an NLR-type immune receptor with a C-terminal PRK domain. The research further reveals the corresponding fungal effector AvrPm6, identified through genome-wide association analysis of Blumeria graminis f. sp. tritici isolates. Biochemical assays demonstrate a direct physical interaction between Pm6 and AvrPm6, which triggers immune activation, reactive oxygen species accumulation, and cell death. Functional validation through gene silencing, overexpression, and gene-editing confirms that Pm6-mediated resistance is strictly dependent on this AvrPm6 recognition. The findings elucidate a previously unknown effector–receptor pair in wheat immunity and provide valuable resources for resistance breeding and engineering of durable disease resistance.
