Electrochemical biosensors based on graphite electrode modified either with copper amine oxidase (EC 1.4.3.6) from grass pea (Lathyrus sativus) or with the amine oxidase coupled with horseradish peroxidase (EC 1.11.1.7) were constructed and used for thedetermination of biogenic amines (histamine, putrescine, cadaverine, tyramine, cystamine, agmatine, spermidine, ethylenediamine, etc.) commonly present in food products. The amine oxidase was effective working both in mono- and bienzyme electrode designsbased either on a direct or mediated electron transfer approach. However the two biosensor designs displayed different selectivity patterns, allowing thus the simultaneous assay of histamine and putrescine. The reaction mechanism of the monoenzymatic unmediated biosensor shows unique features of direct electron transfer achieved for the first time for the copper amine oxidase.
