Gas chromatographic-mas spectrometric determination of low molecular products formed during exposition of polycaprolactone to aspergillus-lipase
Popis výsledku v původním jazyce
This work reports on the determination of possible degradation products released from PCL films aged in the presence of lipase-type enzyme by means of the gas chromatographic-mass spectrometric analysis. The analysis was performed on HP-6890 gas chromatograph equipped with the mass spectrometric detector HP-5673. HP-5MS column (30 m x 0,25 mm x 0,5 mikm, film of bonded 5 % phenyl, 95 % dimethylpolysiloxane, Hewlett-Packard, Waldbronn, Germany) was used. PCL substrate in the film form (Mn = 19 300 g/mol)was added to a phosphate buffer system with pH = 7 containing 0,05 mg ml-1 Aspergillus sp. lipase with the initial enzymatic activity of 0,3 mmol min-1 ml-1. The degradation experiments were carried out for 42 days at 37 °C. For analyses purpose, the aliquots of buffer solution, used in degradation tests, were withdrawn in certain intervals and extracted with dichloromethane. Subsequently, the presence of e-caprolactone was confirmed in the extracts. The decrease in the amount of e-capr
