The research focuses on genotyping Prestice Black-Pied pig (PC) by tetranucleotide microsatellite markers. Genotypes of 575 individuals were determined by a commercial multiplex PCR kit specifically developed for the genotyping of breeding livestock. The kit is recommended for proof of origin according the EU-Directive, kinship testing in context with control of breeding, status of inbreeding for herd book populations. We found further alleles in PC breed outside declared range in six markers (387A12F, S0655, SBH2, SBH10, SBH18, SBH19) out of eleven. In two markers we detected three further alleles, in one case two new alleles and in three markers one allele which we didn't find in other investigated commercial breeds (LW, LA, D, PN). Putative null allele was detected in SBH18 marker. In this locus we observed non- amplified alleles in 16 % from all tested PC individuals. Deviation from Hardy-Weinberg equilibrium (p<0.001) can confirm this hypothesis as well as Hc/H0 difference (Hc 0.836, H0 0.616) - regardless of this marker variability (eleven different alleles in PC breed, PIC is 0.818). However, the tested PC population is not panmictic but small closed population, the divergences can be caused by many reasons such as e.g. selection. The occurrence of null alleles can be very serious problem with interpretation results especially in parentage tests, individual identification, kinship testing and proof of origin. We suggest realize sequence analysis of PCR product for verification the hypothesis of null allele in SBH18 marker in PC breed. The lineage and family analysis are necessary for breeding purposes. Our results provide evidence about differences in genetic structure of PC breed. There are not any information about occurrence of null allele in SBH 18 in PC breed. The other study of null alleles in PC breed can be proposed as a new topic of PC breed research after actually ended project QJ1210253.
