    Bacterial disease is common in pet rabbits, and successful resolution depends on careful and accurate diagnosis, correction of underlying husbandry deficits, and management of concurrent disease processes. Parenteral administration of benzylpenicillin (PNCG) is commonly recommended for the treatment of anaerobic infections and treponematosis2 and in unreasonable dosage intervals (e.g. once in 7 days). However the dosage and treatment regime is not standardized. The aim of this study was to determine PNCG concentrations in rabbit plasma samples after single IM administration (60.000 IU/kg) using an LC-MS/MS validated method with penicillin V as an internal standard (IS). Blood was obtained 12 times within 24 hours. Prior to analysis plasma samples were deproteinised with acetonitrile and penicillins were converted to their piperidine derivatives. The analytes were then separated on a ZORBAX Eclipse XDB C18 (15 cm x 4.6 mm, 5 ?m) column, using a gradient elution program (0.1% formic acid-acetonitrile) followed by mass spectrometry detection in a selective reaction monitoring mode (SRM) on LC-MS/MS system consisting of Agilent 1100 HPLC and Thermo Electron Quantum Discovery Max triple quad mass spectrometer. After 30 min, the PNC concentrations reach their maximal levels (8993.3+-4071.3 ng/ml) and after 12hrs decrease below 126.7+-120.91 ng/ml and after 18hrs to 42.87+-46.30 ng/ml. As MIC of PNCG is in most of the bacteria higher than 60 ng/ml, this drug should be administered twice a day to reach optimal antimicrobial levels
