Maytenus macrocarpa (Ruiz & Pav.) Briq (Celastraceae) is a tree native to the subandean region of the high Amazonian basin. In the local folk medicine (vernacular plant name is ?chuchuhuasha?) the stem and root bark are used for the treatment of rheumatism, diarrhea, bronchitis, fractures and is also valued for its antibacterial, aphrodisiacal and tonic effects. The most used formulation by locals is a decoction or maceration in sugarcane rum. Although chuchuhuasha is among natives very popular, there is still not enough evidences to make it registered evidence-based drug. The most explored compounds of M. macrocarpa species are tetracyclic and pentacyclic triterpenes and dihydro-?-agarofuran sesquiterpenes. Those and other compounds showed through in vitro tests antibacterial, antifungal, antiviral, cytotoxic and anti-inflammatory activity, however overall phytochemical examination of M. macrocarpa is still insufficient. Chloroform portion of ethanolic root bark extract was used for the isolation of tested fractions and subsequently pure constituents. Some extracts showed cytotoxic effect. The non-toxic concentration was used to determination of anti-inflammatory effect. Anti-inflammatory effect was detected by the inhibition of NF-?B/AP-1. For the experiment was used QUANTI-BlueTM medium for detection and quantification of alkaline phosphatase, which was under transcription control of NF-?B/AP-1. Cells were incubated with extracts of concentration 1 ?g/mL and after 1 h the inflammatory response was triggered by bacterial lipopolysaccharide (LPS). After 24 hours of incubation, the activity of NF-?B/AP-1 was evaluated. The ongoing research indicate, some extracts are capable of decreasing of inflammatory effect caused by LPS.
