The easiest technology to produce tablets is direct compression, even though the use of the method requires overcoming many obstacles, mainly related to content uniformity (CU). Some producers offer co-processed excipients (CPE) with various density and particle size distribution. Nowadays, CPE containing commonly processed blends of fillers, binders, disintegrants and other excipients are more and more used. The aim of this work was to assess the effect of CPE physical properties on the CU of resulted tablets. To draw conclusions about mutual relations between predictors variables (CPE physical characteristics) and response variables (CU of tablets), PLS (partial least squares) was employed. The use of CPE appears to be an advantageous option for the formulation of low API tablets by direct compression method. The most important variables influencing CU proved to be a bulk density, pycnometric density and flowability.
