Expansion within the genomic GAA.TTC motif of the first intron of frataxin is connected with the development of Friedreich's ataxia. We observed that p53 in vitro prefers to bind to the (Py-)rich strand over the purine (Pu-)rich strand in non-B DNA substrates formed by sequence derived from the first intron of the frataxin gene. The binding of p53 to this region was confirmed using chromatin immunoprecipitation in humanadenocarcinoma and Friedreich's ataxia fibroblast cells. These observations provide further evidence that p53 binds to non-B DNA structures formed by trinucleotide repeats (TNR), suggesting it is important to investigate further how p53 influences genetic instabilities of TNR sequences.
