In this work of biomedical research were used methods for gene mapping in an experimental mouse model to find loci that control infection with parasites Leishmania major. Leishmania parasites are causative agents of group of diseases called Leishmaniasis which consists of wide spectrum of clinical manifestation from cutaneous to visceral Leishmaniasis. Molecular mechanism of the diseases is mostly unknown that is why there is no effective vaccine against the disease. The main objective of the work was identifying genes/loci controlling pathology in Leishmaniasis caused by L. major series. We conducted our research on F2 hybrids between OcB-43 and O20 mice to map controlling genes. We checked whether our selected genes are involved in response to Leishmania infection, we performed expression of several genes on skin samples from 10 inbreed strains. Statistical analysis showed several novel loci controlling pathology in Leishmaniasis. RT-PCR expression analysis showed that gene expresion depends on genetic background.
