Miniaturized portable high-performance liquid chromatography system for field analysis of hazardous substances
Popis výsledku v původním jazyce
The need for immediate analysis of hazardous substances on-site at the point of sample collection has been ongoing challenge namely for incidents covering accidental environmental pollution, forensic analysis or areas with risk of hazardous materials contamination. Moreover, the field samples are usually complex such that reliable identification and quantitation of target substances is almost impossible without separation of sample components. Liquid chromatography is widely used separation technique for non-volatile samples including drugs, environmental pollutants, toxins, as well as chemical warfare agents or their degradation products and metabolites. Technical development in miniaturization in last decades has opened new possibilities for construction of portable instruments enabling field analysis at the point of need. Herein, we introduce compact miniaturized and portable HPLC instrument (miniLC) equipped with a newly developed combined optical detector capable of broadband spectral detection as well as simultaneous fluorescence monitoring. The system consists of simplified capillary LC scheme based on the single piston pump and selector valve, connected together via capillary which serves as a solvent and sample loop. Selector valve allows automated flow path change and consequent pumping on capillary microcolumn. The detector combines miniature silica capillary detection cell, CCD spectrometer as the light detector, miniature deuterium lamp as the light source for UV/Vis spectra measurement, and LED diode/s for fluorescence excitation. We demonstrate an application of the system for analysis of various hazardous materials, including toxins, organophosphate agrochemicals, carbamates or other CWA surrogates. Presented results demonstrate the capability of miniaturized capillary liquid chromatograph to perform fast (up to 3.5 min) and repeatable separation of agents of concern.
