Advanced Development of Novel Antitubercular Agents Against Multidrug-Resistant Tuberculosis
Anotace anglicky
Tuberculosis (TB) is a highly contagious disease caused by M. tuberculosis (Mtb) each year responsible for more than a million deaths. This makes it the top one cause of deaths caused by a single infectious agent and one of the top causes of deaths worldwide. The most problematic issue with TB is the growing drug resistance of Mtb to the current first- and second-line drugs. This presents a real danger for global population and demands refocusing efforts on the discovery of new potent antitubercular drugs to treat the new drug-resistant cases. This project is focused on advanced experimental development of previously identified low molecular weight compounds with strong in vitro activity against Mtb and non-tuberculous mycobacteria (such as M. avium or M. kansasii), including multidrug-resistant clinical isolates. Described compounds possessed sufficient water solubility and low cytotoxicity against human cells making them excellent starting points for advanced development. Based on simple medicinal chemistry modifications, we aim to further improve their antimycobacterial activity by preparing approximately 40–50 structural analogues to detailly describe the structure-activity relationships for antimycobacterial activity and low cytotoxicity. The most active compounds will then be subjected to advanced microbiological screening including checkerboard studies, time-kill assays, in vivo toxicity determination on G. mellonella model and to preclinical profiling to establish plasma stability and first-phase metabolism. Best selected compounds will undergo in vivo antitubercular activity determination on murine model of TB. Prepared compounds will be pushed forward as new drug candidates for the treatment of drug-resistant tuberculosis or other mycobacterioses prevalent in human population.
