    Barley (Hordeum vulgare) is a key raw material for brewing and food industries, but its quality canbe compromised by Fusarium pathogens responsible for Fusarium Head Blight. Malting is a crucialstep in processing barley, yet Fusarium infection during this phase can negatively impact grainquality and safety. Pulsed electric field (PEF) treatment is a non-thermal microbial control methodsused in food and crop processing, with potential applications in improving malting efficiency.However, the molecular mechanisms underlying barley’s response to both Fusarium infection andPEF treatment during malting remain largelyunexplored.To address this, we investigated transcriptomic changes in Fusarium-infected barley subjected tomalting, with and without PEF treatment. RNA sequencing revealed over 12,000 differentiallyexpressed genes (DEGs) across four malting stages, with the most significant transcriptional shiftsoccurring after 24 hours of germination. Functional enrichment analyses indicated that maltinginduces strong stress responses, including oxidative stress mitigation and hormone-regulated defensemechanisms.PEF-treated barley exhibited fewer DEGs in later stages compared to untreated samples, suggestingthat PEF modulates stress pathways and enhances stability during malting. Key upregulated genes inPEF-treated barley included those linked to abiotic stress resistance, while downregulated geneswere associated with photosynthesis and cell wall remodeling. These findings provide new insightsinto the interplay between malting, pathogen stress, and PEF treatment, highlighting its potential toimprove malting efficiency and barley resilience.
