Cyclodextrins (CDs) are synthesized by the bacterial extracellular enzym cyclodextrin glycosyltransferase (CGTase, E.C. 2.4.1.19) from starch or starch derivatives via an inter- and intramolecural transglycosylation reaction. The production of ?- CD, ß-CD and ?- CD by CGTase from Paenibacillus macerans was studied, in regard to the effect of the source starch (amaranth, corn) on the yield of CDs. The CGTase was produced by a 3-day sterile cultivation in the laboratory Bench-top fermentor BiostatB in acomplex medium containing starch and partially purified by ammonium sulphate precipitation at 60% saturation. Apparent molecular weight as determined by SDS?PAGE was in the range from 105 to 114 kDa. All the commercially important ?-, ß-, and ?-CDs weredetected chromatographically after hydrolysis of the corn and amaranth (Amaranthus cruentus) starch with the isolated enzyme.
