Fish MHC genes - quality vs quantity
Popis výsledku v původním jazyce
Even if the role, variability and evolutionary mechanism of selection of genes of major histocompatibility complex (MHC genes) has been proved for several times to be very important in immunoecological studies and in investigation of associations betweenmultiple parasites and host immunocompetence in cyprinids, the question of quantitative expression of these genes stays still unanswered. Our research reveals that not only ?quality? (genomic variability or allelic richness) but also ?quantity? of thesegenes (active gene expression and its level) may play an important role in host-parasite interactions and may be useful for evolutionary and population studies. By utilizing two modern approaches (high resolution melting analysis ? HRM and quantitativePCR ? qPCR) we obtained notable primary results that could supplement or possibly stand in the analysis by classical methods in MHC research (CE-SSCP, automatic sequencing based on Sanger technology or 454 clonal pyrosequencing).
