Bottom fermenting brewing yeast (S. pastorianus) is a hybrid polyploid microorganism, most often triploid, tetraploid or aneuploid. The hybrid origin of brewing yeast causes problems in identification, including determination of the technologically diverse species S. pastorianus and S. cerevisiae. Wine yeast are mostly diploid and belong to the species S. cerevisiae and S. bayanus. In our study a group of at least 40 brewing, 8 wine and 3 type yeast strains was analyzed by PCR and RFLP techniques. Restriction of mtDNA with 4 different enzymes resulted in 9-12 restriction patterns, an identical pattern was shared by 4 out of 10 ale and lager strain. Wine yeast exhibited a restriction pattern different from that of lager strains. According to the RAPD method, wine and top fermenting strains are clearly separated from lager strains except for 4 top fermenting strains.
