Cryopreservation of common carp Cyprinus carpio and European catfish Silurus glanis spermatozoa for gene resources conservation
Popis výsledku v původním jazyce
Gene resources include original Czech fish strains as a part of our national culture heritage (7 strains of common carp and 2 strains of European catfish). In this study, cryopreservation methods were elaborated for ex situ conservation of Bohemian common carp and European catfish. A sperm freezing procedure was developed, providing good motility of spermatozoa, fertilization and hatching rates in each of the species. Common carp sperm was collected in plastic containers (1/10 - 1/30 volume of sperm andair) and stored under aerobic condition at 4 oC for 1-3 hours. Sperm was diluted 1:5 in a Kurokura extender (128.4 mM NaCl, 2.7 mM KCl, 1.4 mM CaCl2, 2.4 mM NaHCO3) and equilibrated for 40 minutes at 4 oC. Diluted sperm was transferred to 2 ml cryotube sand 10 % of DMSO was added, than the cryotubes were directly transferred to pre-programmed PLANER Kryo 10 series III and cooled from +4 oC to -9 oC at a rate of 4 oC . min-1, then from -11 oC to -80 oC at a rate of 11 oC .min-1, hold for
