The headspace solid-phase microextraction (HS-SPME), coupled to gas chromatography-mass spectrometry (GC-MS), was developed and applied for the detection and identification of volatile compounds in basil (Ocimum basilicum L). Five cultivar types of basil; Purple Opaal, Rot; Trpaslici, Cinamonette and the most common Prava zelena were collected from open experimental garden of the Czech University of Agriculture in Prague. Each cultivar type was grown under organic (compliance to IFOA standard) and conventional conditions. The polydimethylsiloxane/divinylbenzene/carboxene (PDMS-CX-DVB) fibre was proven to be the most suitable for particular purpose since it is able to cover the wide range of analytes present in this type of samples. The most important parameters affecting sorption process such as sample weight, sorption and desorption time and temperature were optimised.
