All retroviruses form a structural polyprotein precursor Gag that directs the assembly of the immature retroviral capsid. The role of nucleic acid in the assembly process was extensively studied and several models predicted how the nucleic acid could work and participate on retroviral capsid assembly, but still the exact function of nucleic acid is not clear. We have studied the in vitro assembly process of Mason-Pfizer monkey virus Gag deletion mutant, consisting of two structural proteins, capsid andnucleocapsid. We have focused mainly on (pro-)CANC, CA-NC fusion protein lacking its N-terminal proline, aminoacid that is determining the shape of assembled particles. This mutant assembles with high efficiency in vitro, forming spherical particles of 75 nm diameter in average.To study the role of nucleic acids in the assembly process we optimized the protein purification method that provides the nucleic acid free protein.
